  The major hormone derived from the thyroid gland. Thyroxine is synthesized via the iodination of tyrosine (monoiodotyrosine) and the coupling of iodotyrosine (diiodotyrosine) in thyroglobulin. Thyroxine is separated from thyroglobulin by proteolysis and released in the blood. Thyroxine is peripherally deiodinated to form triiodothyronine which exerts broad stimulatory effects on cell metabolism.

The major form of thyroid hormone in the blood is T4. The ratio of T4 to T3 is roughly 20 to 1. Thyroxine is converted to active T3 (three to four times more potent than T4) within cells by deiodinases (5'-iodinase). These are further processed by decarboxylated and deiodinated to produce iodothyronamine (T1 a) and thyronamine (T0 a).
