Description

The complement fixation test is used to determine the presence of antigens or antibodies in a patient specimen. The patient specimen must first have the innate complement inactivated by heating it. Then, antigen (or antibody if looking for an antigen) and complement of known concentrations are added to the specimen. Antibodies in the patient specimen are allowed to react with the antigen. Complement will be fixed if antigens have bound to antibodies. To visualize the reaction, sheep red blood cells and antibodies against sheep red blood cells are added to the reaction vessel. If an antigen-antibody reaction has occurred, the complement will have been fixed and the sheep cells will not lyse.
