Term Description
The stain is interpreted as positive if 50% or more of the tumor cells show moderate to strong (2+, 3+) membranous staining, indicating MET amplification. MET amplification (positive) has been seen in approximately 22% of non-small-cell lung cases with acquired resistance to anti-EGFR inhibitors. Recent Phase II clinical trails (OAM4558g) results were released at ASCO in 2011 showed that patients who were c-MET positive (2+ or 3+) lived twice as long without disease progression (PFS) when they received MetMAb therapy in combination with erlotinib versus receiving erlotinib alone.

Hepatocyte growth factor receptor (HGFR, also known as mesenchymal-epithelial transition (MET or c-MET]) is a transmembrane tyrosine kinase receptor that transduces signals from the extracellular matrix into the cytoplasm by binding to hepatocyte growth factor/HGF ligand. [UniProt: P08581] The HGFR is a product of the MET gene and is important to oncogenesis and normal cellular function. It regulates many physiological processes including proliferation, scattering, morphogenesis and survival. The HGFR can be overexpressed in epithelial cancers, such as lung cancer. The overexpression of c-MET is thought to allow for cell migration and invasion leading to metastasis as well as growth advantage. PMID: 24005867
